Abstract:
Objective To investigate the feasibility of multi-slice spiral CT (MSCT)examination in animal silicosis model, the MSCT examination results were compared with the pathological examination result in rat silicosis model.
Methods Sixty rats without specific pathogen grade were divided into control group, silicosis model group and hUC-MSCs treatment group. After 60 days, all rats were examined by MSCT and finally received pathological examination. The MSCT classification of rat silicosis model was made according to the small shadow density of each lung window. The pathological grades of the rat silicosis model were determined according to the percentage of the area of the inflammatory granuloma, silicotic nodule and silicotic fibrosis in the lung tissue. The MSCT classification and pathological grading in different groups of rats with silicosis were compared by nonparametric test (Kruskal-Wallis test); the consistency of MSCT classification and pathological grading in rat silicosis model was tested by weighted Kappa consistency test.
Results Both MSCT classification (H=26.27, P < 0.01)and pathological grade (H=38.86, P < 0.01) showed that there were statistically significant differences among different groups. The consistency test (Kappa=0.706, P < 0.05) suggested that the MSCT classification and pathological grade in rat silicosis model were higher consistent.
Conclusion MSCT examination, as a non-fatal examination, can observe the development of silicosis stage in rat silicosis model, and it can be used as an examination method to track the effect of stem cell treatment on rat silicosis model.
